The Department of Health says more than 16 000 people have received Lenacapavir since the rollout of the groundbreaking HIV prevention injection began in June.
The long acting medicine is given just twice a year and is designed to prevent HIV infection before exposure.
South Africa launched the rollout after receiving an initial shipment of nearly 38 000 doses, with priority groups including young women, sex workers and other people at higher risk of HIV.
Health Minister Aaron Motsoeledi officials say the strong early uptake shows communities are embracing the new prevention option as the country works to reduce new HIV infections by 2030.

The Robin Hood Foundation helped bring warmth and companionship to vulnerable elderly residents in Embo near Hillcrest.
Working with Pastor Leonard and the Power of God Community Church, volunteers spent the day sharing meals, conversation, music and laughter with around 55 senior citizens who meet every month for fellowship.
The foundation also handed out care packages containing pre loved clothing, linen, maize meal and winter essentials including blankets, beanies and gloves.

Conservation organisation Elephants Alive has received a R79 000 donation from Hot Cares to help protect elephants and nearby communities along the western boundary of the Kruger National Park.
The funding will pay for a new satellite tracking collar and veterinary support, allowing conservation teams to monitor elephant movements and prevent herds from entering neighbouring villages.
Recent flooding in the park disrupted traditional migration routes, increasing the risk of human elephant conflict. Elephants Alive also uses non lethal measures such as beehive fences, chilli deterrents and Elephant Shepherd teams to safely guide animals away from communities.
